目的:建立复方替硝唑含漱液中替硝唑和醋酸氯己定同时测定含量的高效液相检测方法。方法:色谱柱为AgilentZORBAX Eclipse XDB-C_(18)(250 mm×4.6 mm,5μm);以甲醇-乙腈-三乙胺缓冲液(0.05 mol·L(-1)用磷酸调pH至3.0)(45:15:40)为流动相;检测波长为260 nm;流速为1.0 ml·min~(-1);柱温为30℃。结果:替硝唑和醋酸氯己定的线性范围分别为25.17~251.7μg·ml~(-1)(r=0.999 9)和10.11~101.1μg·ml~(-1)(r=0.999 8);平均回收率分别为97.90%(RSD=0.93%,n=6)和98.19%(RSD=0.56%,n=6)。结论:该方法简便快速,结果准确可靠,可作为复方替硝唑含漱液的质量控制方法。
Objective: To establish a HPLC method for the simultaneous determination of tinidazole and chlorhexidine acetate in tinidazole gargle. METHODS: The column was Agilent ZORBAX Eclipse XDB-C 18 (250 mm × 4.6 mm, 5 μm). The pH was adjusted to 3.0 with 0.05 mol·L -1 phosphate in methanol-acetonitrile-triethylamine buffer 45:15:40) as the mobile phase; the detection wavelength was 260 nm; the flow rate was 1.0 ml · min -1; the column temperature was 30 ℃. Results: The linear ranges of tinidazole and chlorhexidine acetate were 25.17 ~ 251.7μg · ml ~ (-1) (r = 0.999 9) and 10.11 ~ 101.1μg · ml ~ (-1) (r = 0.999 8) The average recoveries were 97.90% (RSD = 0.93%, n = 6) and 98.19% (RSD = 0.56%, n = 6), respectively. Conclusion: The method is simple, rapid and accurate. It can be used as a quality control method for compound tinidazole gargle.
