从雷公藤(Tripterygium Wilfordii Hook.f.)的根皮中提取的雷公藤内酯醇,对小鼠白血病和人的离体白血病细胞都有抗癌活性。本实验的目的是探讨此药物的作用原理。一、陷窝细胞是小肠上皮组织的干细胞,常被用来研究抗癌药物的体内作用机理。小鼠腹腔内一次注射雷公藤内酯醇(0.1r/每g体重)后,3小时出现凋落(apoptosis),5小时更为显著。光镜放射自显影证实凋落可出现于受[~3H]TdR标记或未受标记的细胞,提示此药物可作用于S期和非S期细胞,为周期非特异性药物。据文献,凋落有别于一般凝固性坏死,可见于许多生理和病理情况下,多种抗癌药均可引起,故属非特异性病变。
Triptolide extracted from the root bark of Tripterygium Wilfordii Hook.f. has anticancer activity against murine leukemia and ex vivo human leukemia cells. The purpose of this experiment is to investigate the principle of action of this drug. First, the pit cells are the stem cells of the small intestine epithelium. They are often used to study the in vivo mechanism of anticancer drugs. After mice were intraperitoneally injected with triptolide (0.1 r/g body weight), there was apoptosis at 3 hours, which was more pronounced at 5 hours. Light microscopy autoradiography confirmed that litter can occur in [~3H]TdR-labeled or unlabeled cells, suggesting that the drug can act on S-phase and non-S-phase cells, and is a non-specific drug for the cycle. According to the literature, litterfall is different from general coagulative necrosis. It can be found in many physiological and pathological conditions. Many anticancer drugs can cause it, so it is a non-specific lesion.
