目的分析5种阳性致敏药物对小鼠耳后淋巴结7种免疫细胞表面分子表达和6个细胞亚型百分比的变化,探讨药物致敏中淋巴结特意免疫细胞表型的变化。方法选用24只健康小鼠,每组4只,在本实验室驯化1周后分别使用致敏药物耳后涂抹刺激后,提取耳后淋巴结细胞,使用流式细胞仪检测CD45+CD3+%,CD3+CD4+%,CD4+CD154+%,CD4+CD28+%,CD45+CD19+%和CD45+CD69+%细胞亚群的百分比情况。结果丁子香酚和2-己基肉桂醛致敏后7种表面分子表达与空白对照组相应表型表达出现明显差异;戊二醛、2,4-二硝基氯苯和巯基苯并噻唑致敏后出现部分表达的差异性变化。结论可以初步判断CD4和CD28的表达可能参与了致敏效应,可能成为初筛药物致敏效应的细胞表型。
OBJECTIVE: To analyze the changes of the surface molecules expression and the percentage of 6 cell subtypes of 7 kinds of immune cells in the posterior ear lymph nodes of 5 kinds of positive sensitized drugs, and to explore the changes of the phenotypes of the specific immune cells in drug-sensitized lymph nodes. Methods Twenty-four healthy mice were selected, 4 rats in each group. After being acclimated for 1 week in our laboratory, the sensory drugs were smeared on the ear and the post-ear lymph node cells were extracted. Flow cytometry was used to detect CD45 + CD3 +, CD3 + CD4 +%, CD4 + CD154 +%, CD4 + CD28 +%, CD45 + CD19 +% and CD45 + CD69 +% cell subsets. Results After the eugenol and 2-hexylcinnamaldehyde were sensitized, the expression of seven kinds of surface molecules was significantly different from that of the blank control group. Glutaraldehyde, 2,4-dinitrochlorobenzene and mercaptobenzothiazole sensitized After the emergence of some of the differences in expression. Conclusion It can be preliminarily determined that the expression of CD4 and CD28 may be involved in the sensitization effect and may become the cell phenotype of the initial screening drug sensitization effect.
