目的:研究Hsp70在TLR信号途径中对肿瘤细胞增殖的影响。方法:将体外培养的H22细胞分成对照组和用Hsp70刺激的实验组,设计基因表达的引物、PCR扩增、RT-PCR检测两组细胞的TLR2和TLR4的mRNA表达水平,Western blot检测β-catenin水平,对培养10天后的H22细胞进行计数。结果:Hsp70分别处理H22细胞8h后,基因TLR2、TLR4mRNA的表达均明显上调;Hsp70促进H22细胞中β-catenin上调表达;Hsp70处理组的细胞增殖倍数高于对照组。结论:Hsp70在TLR信号途径中具有促进肿瘤细胞增殖作用。
Objective: To investigate the effect of Hsp70 on tumor cell proliferation in the TLR signaling pathway. Methods: H22 cells cultured in vitro were divided into control group and experimental group stimulated by Hsp70. The gene expression primers were designed. The mRNA expression levels of TLR2 and TLR4 were detected by PCR and RT-PCR. catenin levels, counting of H22 cells after 10 days of culture. Results: After treated with Hsp70 for 8h, the expression of TLR2 and TLR4 mRNA were significantly up-regulated. Hsp70 promoted the up-regulation of β-catenin in H22 cells. The proliferation of Hsp70-treated group was higher than that of control group. Conclusion: Hsp70 can promote tumor cell proliferation in TLR signaling pathway.
