目的:探讨祛痰化瘀通脉颗粒对急性心肌缺血大鼠血管新生的影响及其机制。方法:对大鼠行左冠状动脉前降支结扎术致急性心肌梗死模型,术后1至5d口服灌胃给予大、中、小剂量祛痰化瘀通脉颗粒,每日1次。采用伊文氏蓝-氯化三苯基四唑(TTC)双重染色法测定心肌缺血和心肌坏死面积,用免疫组化法测定缺血心肌微血管密度(MVD),检测心梗边缘区血管内皮生长因子(VEGF)及VEGFR-2的表达,观察不同剂量祛痰化瘀通脉颗粒对心肌缺血大鼠缺血心肌血管新生的影响。结果:各剂量祛痰化瘀通脉颗粒组术后可明显缩小心肌梗死及缺血面积,明显增加新生血管密度,VEGF、VEGFR-2表达均有明显升高,与模型组比较,差异均有统计学意义(P<0.05)。结论:祛痰化瘀通脉颗粒能促进心肌梗死后大鼠缺血心肌血管新生,增加微血管密度,其作用机制可能与与上调心肌局部VEGF、VEGF-2相关。
Objective: To investigate the effect of Qutan Huayu Tongmai Granule on angiogenesis in acute myocardial ischemia rats and its mechanism. Methods: Acute myocardial infarction model was induced in rats by ligation of the left anterior descending coronary artery. Large, medium and small doses of Qutan Huayu Tongmai Granule were administered orally once to 5 days after operation. The areas of myocardial ischemia and myocardial necrosis were determined by double staining of Evans blue and triphenyltetrazolium chloride (TTC). The myocardial microvascular density (MVD) was measured by immunohistochemistry and the vascular endothelial growth (VEGF) and VEGFR-2 expression in rats to observe the effect of different doses of Qutan Huayu Tongmai Granule on ischemic myocardial angiogenesis in rats with myocardial ischemia. Results: Expectorant stasis, Tongyu granule group could obviously reduce myocardial infarction and ischemic area, significantly increase neovascular density, and significantly increase the expression of VEGF and VEGFR-2. Compared with the model group, the difference was Statistical significance (P <0.05). Conclusion: Qutan Huayu Tongmai Granule can promote angiogenesis and increase microvessel density in ischemic myocardium of rats after myocardial infarction, and its mechanism may be related to up-regulation of local VEGF and VEGF-2.
