目的:通过体外T淋巴细胞增殖实验筛选过山枫抗炎作用的活性部位;对确定的活性部位进一步探讨对T淋巴细胞凋亡的影响。方法:对过山枫乙醇提取物进行分离提取,采用体外小鼠脾淋巴细胞增殖实验,检测过山枫不同提取部位对小鼠脾细胞增殖的影响,从中筛选出对小鼠脾细胞增殖有抑制作用的活性部位。采用流式细胞技术观察活性部位对小鼠脾淋巴细胞凋亡的影响。结果:过山枫不同极性部位及从过山枫乙酸乙酯部位(GSF-A)分离得到3个组分对体外培养的T淋巴细胞增殖均有不同程度的抑制作用;GSF-A 3个活性部位对T淋巴细胞凋亡有显著促进作用。结论:过山枫不同极性部位均具有抗炎活性,且GSF-A的活性显著;从GSF-A部位中分离得到的3个部位均具有抗炎活性;过山枫对T淋巴细胞增殖的抑制及诱导凋亡的作用一致,提示诱导凋亡是过山枫抑制T淋巴细胞增殖的机制之一。
OBJECTIVE: To screen the active sites of the anti-inflammatory effects of T. mongolica through in vitro T-lymphocyte proliferation assays, and further explore the effects of the identified active sites on the apoptosis of T lymphocytes. METHODS: The ethanol extracts were extracted and used for the extraction of mouse spleen lymphocytes in vitro to test the effect of different extraction sites of T.mauriculata on the proliferation of mouse spleen cells, and the inhibition of the proliferation of mouse spleen cells was selected. RESULTS: Three fractions isolated from different polar parts of T. cinnamomea and isolated from ethyl acetate (GSF-A) had inhibitory effects on the proliferation of T lymphocytes cultured in vitro; GSF-A 3 The active site has a significant role in promoting the apoptosis of T lymphocytes. Conclusion: All polar parts of A. transcarpa have anti-inflammatory activity, and the activity of GSF-A is significant; all three sites isolated from GSF-A site have anti-inflammatory activity; The inhibition and induction of apoptosis are consistent, suggesting that inducing apoptosis is one of the mechanisms by which T. mongolica inhibits the proliferation of T lymphocytes.
