目的:探讨清心开窍方有效成分对AD大鼠学习记忆能力及脑组织细胞凋亡基因Bax、Bcl-2 mRNA及胶质纤维酸性蛋白(GFAP)mRNA表达的影响。方法:动物实验采用对照观察法。雄性SD大鼠56只,随机分为正常组、模型组、安理申组、清心开窍方组、皂苷组、多糖组和挥发油组,每组8只。采用双侧杏仁核注射Aβ25-35制造AD大鼠模型,造模后第2天起,分别灌胃给药安理申、清心开窍方水煎剂、皂苷、多糖、挥发油及双蒸水,2周后,观察大鼠水迷宫空间记忆能力,并通过RT-PCR研究脑组织Bax、Bcl-2 mRNA及GFAP mRNA的表达。结果:中药各组能明显改善AD大鼠学习记忆能力,大鼠大脑皮层、海马区Bax、GFAP mRNA表达水平降低,清心开窍方组、皂苷组皮质Bcl-2 mRNA表达水平升高,与模型组比较差异显著(P<0.05,P<0.01)。结论:清心开窍方能显著改善AD大鼠学习记忆的能力,可能是通过降低皮质、海马Bax mRNA及GFAP mRNA表达,升高Bcl-2 mRNA表达发挥作用。
Objective: To investigate the effects of the active ingredients of “Qingxin Kaiqiao Recipe” on the learning and memory abilities and the expressions of Bax, Bcl-2 mRNA and glial fibrillary acidic protein (GFAP) mRNA in brain of AD rats. Methods: Animal experiments using the control method. Fifty-six male Sprague-Dawley rats were randomly divided into normal group, model group, Amoxicillin group, Qingxin Kaiqiao Recipe group, saponin group, polysaccharide group and volatile oil group. AD rats were injected with Aβ25-35 by bilateral amygdala. After 2 days, the rats were orally administered with Amoxicillin, Qingxin Kaiqiao decoction, saponin, polysaccharides, volatile oil and double distilled water for 2 weeks After that, the spatial memory ability of water maze was observed, and the expression of Bax, Bcl-2 mRNA and GFAP mRNA in brain tissue were detected by RT-PCR. Results: The expressions of Bax and GFAP mRNA in cerebral cortex and hippocampus decreased significantly in all groups, and the expression of Bcl-2 mRNA in cortex of Qixin Kaiqiao Recipe group and saponin group was significantly increased. Compared with model group The difference was significant (P <0.05, P <0.01). Conclusion: Qixin Kaiqiao Recipe can significantly improve the learning and memory ability of AD rats, which may be through decreasing the expression of Bax mRNA and GFAP mRNA in cortex and hippocampus and increasing the expression of Bcl-2 mRNA.
