质粒标准分子是指含有外源基因和内源标准基因特异性片段的重组质粒分子。DNA条形码技术是通过对标准目的基因的DNA序列进行分析从而进行物种鉴定的技术。构建基于DNA条形码的质粒标准分子是DNA条形码技术应用于检测实践的要求。本研究将这两种检测鉴定技术相结合应用于检疫性疫霉的检测,构建了11种检疫性疫霉的DNA条形码标准分子,进行了测序验证、均匀性、稳定性和特异性验证。结果表明,构建的质粒标准分子准确度、均匀性、稳定性和特异性均良好,对实际口岸检验检疫工作具有实践应用价值。
Plasmid standard molecules refer to recombinant plasmid molecules containing exogenous genes and specific fragments of endogenous standard genes. DNA barcoding is a technique for species identification by analyzing the DNA sequence of a standard target gene. Construction of DNA-based barcode-based plasmid standard molecules is the application of DNA barcode technology to test practice requirements. In this study, two kinds of detection and identification techniques were applied to the detection of quarantine Phytophthora sojae, and 11 DNA barcode standard molecules of quarantine Phytophthora infestans were constructed and verified by sequencing, homogeneity, stability and specificity. The results showed that the construction of plasmid standard molecules accuracy, uniformity, stability and specificity are good, practical inspection and quarantine of the actual port has practical value.
