目的探讨外侧下丘脑orexin能神经元区微注射腺苷对SD大鼠睡眠-觉醒周期的影响。方法选用成年雄性SD大鼠14只,按3种不同的给药模式在外侧下丘脑orexin能神经元区分别微注射1、10、20 nmol/μl腺苷,以人工脑脊液为自身对照,然后采用脑电机同步记录大鼠的脑电和肌电活动,观察外侧下丘脑区微注射腺苷后大鼠睡眠-觉醒周期的变化。结果与人工脑脊液自身对照相比,在大鼠外侧下丘脑分别微注射1、10 nmol/μl和20 nmol/μl腺苷后的3 h内,大鼠觉醒时间明显减少到人工脑脊液自身对照的84%、62%和60%,并且大鼠睡眠时间均有不同程度的延长(P<0.05)。在微注射20 nmol/μl腺苷后,大鼠觉醒时间持续减少了3 h,并且与微注射1、10 nmol/μl腺苷后相比,其觉醒时间的减少和NREM/REM睡眠时间的增加也最明显。结论在外侧下丘脑orexin能神经元区微注射腺苷促进大鼠睡眠。
Objective To investigate the effect of microinjection of adenosine on orexin neurons in lateral hypothalamus on sleep-wake cycle in SD rats. Methods Fourteen adult male Sprague-Dawley rats were randomly divided into 3 groups: 1, 10, 20 nmol / μl adenosine, and artificial cerebrospinal fluid The brain electrical and motor activities of rats were synchronously recorded, and the changes of sleep-wake cycle in rats after hypothalamic microinjection of adenosine were observed.
