目的研究蛋氨酸脑啡肽抗肿瘤作用及对SH-SY5Y细胞内钙离子浓度影响。方法用CCK-8法,检测蛋氨酸脑啡肽对SH-SY5Y细胞的生长抑制作用;显微镜下记录给药前后细胞生长情况;流式细胞术结合Fluo 4-AM染色法,检测细胞内钙离子浓度变化,用SAS 9.1统计分析。结果蛋氨酸脑啡肽可通过降低细胞内钙离子浓度对SH-SY5Y细胞生长产生浓度特异性抑制作用。结论细胞内钙离子浓度变化参与蛋氨酸脑啡肽抑制SH-SY5Y肿瘤细胞的生长。
Objective To study the antitumor effect of enkephalin and its effect on the intracellular calcium concentration in SH-SY5Y cells. Methods The growth inhibition of SH-SY5Y cells by methionine enkephalin was detected by CCK-8 method. The growth of SH-SY5Y cells was observed under microscope. The intracellular calcium concentration was detected by flow cytometry and Fluo 4-AM staining Change, using SAS 9.1 statistical analysis. Results Methionine enkephalin can inhibit the growth of SH-SY5Y cells in a concentration-dependent manner by decreasing the intracellular calcium concentration. Conclusion The changes of intracellular calcium concentration are involved in the methionine enkephalin inhibition of SH-SY5Y tumor cell growth.
