目的观察冬凌草甲素对人乳腺癌细胞株Bcap-37细胞的生长抑制作用,初步探讨冬凌草甲素调控乳腺癌细胞增殖的分子机制。方法 MTT比色法检测冬凌草甲素对Bcap-37细胞的增殖抑制作用,相差显微镜观察细胞形态变化,流式细胞仪分析细胞周期分布,RT-PCR检测p21、cdc2和cyclin B1 mRNA表达。结果冬凌草甲素对Bcap-37细胞有显著的增殖抑制作用,呈时间剂量依赖趋势(P<0.01);处理后的Bcap-37细胞发生明显的形态改变;作用24h后,Bcap-37细胞在G2/M期阻滞;冬凌草甲素作用Bcap-37细胞后,p21 mRNA水平的表达增加,而cdc2、cyclin B1 mRNA的表达降低(均P<0.05)。结论冬凌草甲素可抑制乳腺癌细胞的增殖,该作用可能与p21 mRNA表达上调及cdc2、cyclin B1 mRNA表达下调有关。
Objective To observe the inhibitory effect of oridonin on the growth of human breast cancer cell line Bcap-37 and to investigate the molecular mechanism of oridonin in regulating the proliferation of breast cancer cells. Methods MTT assay was used to detect the inhibitory effect of oridonin on the proliferation of Bcap-37 cells. The morphological changes of cells were observed by phase contrast microscopy. The cell cycle distribution was analyzed by flow cytometry. The mRNA expressions of p21, cdc2 and cyclin B1 were detected by RT-PCR. Results Oridonin could significantly inhibit the proliferation of Bcap-37 cells in a dose-and-time-dependent manner (P <0.01). The morphological changes of Bcap-37 cells were observed after treated with oridonin for 24 hours, After G2 / M arrest, the expression of p21 mRNA increased and the expression of cdc2 and cyclin B1 mRNA decreased after treated with oridonin (all P <0.05). Conclusion Oridonin can inhibit the proliferation of breast cancer cells, which may be related to the up-regulation of p21 mRNA and the down-regulation of cdc2 and cyclin B1 mRNA expression.
