采用主成分分析法(PCA)和偏最小二乘法(PLS)对太白美花草归经规律进行研究。同时建立高效液相色谱-三重四级杆-电喷雾离子源串联质谱法(HPLC-ESI-MS)同时测定13种毛茛科药材中55种活性成分的定量分析方法。该研究采用Agilent 5 TC-C18(2)色谱柱(4.6 mm×250 mm,5μm),以含0.1%甲酸的乙腈-水为流动相梯度洗脱,HPLC-ESI-MS正负离子模式下,以多反应监测(MRM)扫描采集数据,外标法定量。采用PCA、PLS-DA法对毛茛科13种代表药材进行多元统计分析,以评价太白美花草和这些药材在性味、归经方面的相似性和差异性。分析结果显示55种化合物线性关系良好,r?0.99,精密度、重复性和稳定性良好。PCA结果与PLS分析结果判定太白美化草归经为肺、膀胱经,味辛、性温。说明本研究建立的多种小分子化合物的定量分析以及PLS方法适用于发掘中药的性味归经规律,可以作为珍稀草药替代性研究的方法。
Principal component analysis (PCA) and partial least squares (PLS) were used to study the law of the transpiration of. Simultaneous simultaneous determination of 55 active constituents in 13 Ranunculaceae by HPLC-ESI-MS with triple quadrupole-electrospray ionization mass spectrometry (ESI-MS / MS) was established. Using the method of PCA and PLS-DA, the multivariate statistical analysis of 13 representative species of Ranunculaceae was carried out to evaluate the similarities and differences of the herbs and the herbs in this field. The analytical results showed that the 55 compounds showed a good linear relationship, r? 0.99, with good precision, reproducibility and stability. PCA results and PLS analysis results judged Taibai beautification of grass for the lung, bladder, spicy, warm. The quantitative analysis of a number of small molecule compounds established in this study and the application of PLS method to explore the law of taste and taste of traditional Chinese medicine can be used as alternative methods for the research of rare herbs.
