研究rt-PA工程细胞在小型填充床反应器中连续高密度培养的过程,以及在连续灌注培养条件下rt-PA表达水平、葡萄糖的消耗率、乳酸和氨与rt-PA表达水平之间的关系以探索该工程细胞培养的最佳条件。在不中断培养的情况取样作细胞计数、rt-PA和代谢物的检测。结果:rt-PA工程细胞在小型填充床反应器中最高细胞数可达3.8×109细胞,最高rt-PA浓度达12mg/L结论:这种新型的小型填充床反应器是操作方便、利用价值高的研究设备。
