【目的】研究中药益气血方对超促排卵小鼠卵巢生长分化因子-9(growth differentiation factor-9,GDF-9)与GDF-9B表达的影响。【方法】选用昆明种小鼠接受促性腺激素释放激素类似物(GnRHa)与孕马血清(PMSG)超促排卵,中药组予以益气血方(经后增殖颗粒)(剂量为1.5 mg/kg)灌胃,对照组予以生理盐水灌胃。注射绒毛膜促性腺激素(hCG)后取卵巢组织,采用免疫组织化学方法检测GDF-9与GDF-9B的表达与半定量分析,采用实时荧光定量PCR方法检测GDF-9、GDF-9B mRNA的含量。【结果】卵母细胞与颗粒细胞中检测到GDF-9的表达,GDF-9B未表达于卵母细胞,而在颗粒细胞中高表达。半定量分析结果显示中药组卵巢组织中GDF-9与GDF-9B蛋白表达均显著高于对照组(P<0.05),实时荧光定量PCR结果显示中药组GDF-9、GDF-9B mRNA表达水平均显著高于对照组(P<0.05)。【结论】中药益气血方可促进超促排卵小鼠卵巢GDF-9与GDF-9B的表达,可能通过此机制促进卵母细胞发育潜能,从而促进胚胎种植。
【Objective】 To investigate the effect of Yiqi Xuefang on the expression of ovary growth differentiation factor-9 (GDF-9) and GDF-9B in ovariectomized mice. 【Method】 Kunming mice were treated with GnRHa and PMSG for ovulation induction. The Chinese medicine group was given Yiqixue Formula (post-proliferative granules) (1.5 mg / kg ) Gavage, the control group was given saline. Ovarian tissues were obtained after injection of human chorionic gonadotropin (hCG), the expression and semi-quantitative analysis of GDF-9 and GDF-9B were detected by immunohistochemistry, and the expressions of GDF-9 and GDF-9B mRNA were detected by real-time fluorescence quantitative PCR content. 【Results】 The expression of GDF-9 was detected in oocytes and granulosa cells. GDF-9B was not expressed in oocytes but highly expressed in granulosa cells. The results of semi-quantitative analysis showed that the expression of GDF-9 and GDF-9B in ovarian tissues of Chinese medicine group were significantly higher than those in control group (P <0.05). The real-time quantitative PCR results showed that the expressions of GDF-9 and GDF- Significantly higher than the control group (P <0.05). 【Conclusion】 Yiqi Xue can promote the expression of GDF-9 and GDF-9B in the ovary of ovariectomized mice, which may promote oocyte developmental potential and promote embryo implantation.
