目的 :探讨miR-181a在唾液腺腺样囊性癌细胞侵袭、转移中的作用。方法 :利用QRT-PCR检测miR-181a在一对不同侵袭、迁移能力细胞株中的表达水平。过表达或沉默miR-181a在SACC-LM和SACC-83细胞中的表达。利用激光共聚焦显微镜观察转染后细胞骨架的改变。Western免疫印迹检测转染后侵袭、转移相关因子的表达。通过尾静脉注射miR-181a mimics和mimics NC细胞,建立裸鼠肺转移模型。采用SPSS17.0软件包对数据进行统计学处理。结果:QRT-PCR结果显示,miR-181a在低侵袭、迁移能力SACC-83细胞中的表达量显著高于高侵袭、迁移能力的SACC-LM细胞。SACC-LM细胞转染miR-181a mimics后,细胞呈梭形,Slug、p ERK1/2、ERK1/2表达水平下调;SACC-83细胞转染miR-181a LNA后,细胞变圆,体积变小,Slug、p ERK1/2、ERK1/2表达水平升高。裸鼠肺转移模型miR-181a mimics组,细胞形成肺转移灶面积显著小于mimics NC组(P<0.05)。结论 :miR-181a能抑制SACC的侵袭和转移。
Objective: To investigate the role of miR-181a in invasion and metastasis of salivary adenoid cystic carcinoma cells. Methods: The expression of miR-181a in a pair of different invasion and migration cell lines was detected by QRT-PCR. Overexpression or silencing of miR-181a expression in SACC-LM and SACC-83 cells. The change of cytoskeleton after transfection was observed by laser confocal microscopy. Western blotting was used to detect the expression of invasion and metastasis-related factors after transfection. The lung metastasis model of nude mice was established by injecting miR-181a mimics and mimics NC cells through tail vein. SPSS17.0 software package for statistical analysis of the data. Results: The results of QRT-PCR showed that the expression of miR-181a in SACC-83 cells with low invasion and migration ability was significantly higher than that in SACC-LM cells with high invasion and migration ability. SACC-LM cells transfected with miR-181a mimics showed spindle-shaped cells, decreased expression of Slug, p ERK1 / 2 and ERK1 / 2. After transfected with miR-181a LNA, the cells became round and the volume became smaller , Slug, p ERK1 / 2, ERK1 / 2 expression increased. In miR-181a mimics group, the lung metastasis area of lung metastasis model in nude mice was significantly smaller than that in mimics NC group (P <0.05). Conclusion: miR-181a can inhibit the invasion and metastasis of SACC.
