目的构建针对胶质瘤发生、发展分子机制研究的组织芯片。方法根据研究目的设计118例样本(包括各级别人胶质瘤组织、恶性胶质瘤细胞系多细胞球体、胶质瘤干细胞球体以及用于对照的正常人脑组织、人胚胎脑组织、神经干细胞球体)的阵列,应用组织芯片仪制作组织芯片蜡块,切片后进行HE染色和光镜观察。结果构建的组织芯片蜡块包含118个点阵;熔蜡后有个别点阵发生移位,切片后无点阵缺失;HE染色观察符合原始病理诊断。结论针对胶质瘤发生、发展分子机制研究而制作的组织芯片是胶质瘤分子病因研究的有用工具。
Objective To construct tissue microarray for the study of molecular mechanism of glioma development and progression. Methods According to the purpose of the study, 118 samples were designed, including glioma tissues of various grades, glioma cell lines of multicellular spheroids, glioma stem cell spheres and normal human brain tissues, human embryonic brain tissues, neural stem cells Sphere) array, the application of tissue microarray tissue chip wax, sliced HE staining and light microscopy. Results The tissue chip wax block consisted of 118 dots. After the wax was melted, some dots were displaced and no lattice defects were observed after slicing. HE staining was consistent with the original pathological diagnosis. Conclusion Tissue microarray designed according to the molecular mechanism of glioma development and development is a useful tool to study the molecular etiology of glioma.
