目的:建立同时测定复方磺胺甲唑片中磺胺甲唑和甲氧苄啶含量的高效液相色谱法。方法:采用KromasilC18色谱柱(150mm×4.6mm,5μm),甲醇-水-冰醋酸-三乙胺(225∶345∶0.5∶0.2)为流动相,流速为0.7ml/min,检测波长为239nm。结果:结果磺胺甲唑在15~75μg/ml范围内,峰面积与其浓度线性关系良好(r=0.9994),平均回收率为99.4%,重复性实验RSD为0.8%(n=6),检测限为0.1μg/ml;甲氧苄啶在3~15μg/ml的范围内,峰面积与其浓度线性关系良好(r=0.9992),平均回收率为99.1%,重复性试验RSD为0.6%(n=6),检测限为0.02μg/ml。结论:该法准确、简便、快速、专属性强,灵敏度高,适用于复方磺胺甲唑片的含量测定。
Objective: To establish a HPLC method for simultaneous determination of sulfamethoxazole and trimethoprim in compound sulfamethoxazole tablets. Methods: A Kromasil C18 column (150mm×4.6mm, 5μm) was used. The mobile phase consisted of methanol-water-glacial acetic acid-triethylamine (225∶345∶0.5∶0.2). The flow rate was 0.7ml/min. The detection wavelength was 239nm. RESULTS: The results showed that the sulfamethoxazole was in the range of 15-75 μg/ml, and the linear relationship between the peak area and the concentration was good (r=0.9994). The average recovery was 99.4%. RSD was 0.8% (n=6) for the repeatability test. The limit was 0.1 μg/ml; trimethoprim ranged from 3 to 15 μg/ml, and the linear relationship between peak area and concentration was good (r=0.9992), the average recovery was 99.1%, RSD was 0.6% for repeatability test (n = 6), the detection limit was 0.02 μg/ml. Conclusion: The method is accurate, simple, rapid, specific and high sensitivity. It is suitable for the determination of compound sulfamethoxazole tablets.
