目的:探讨乳腺癌组织中Skp2与p27Kip1的表达及其意义。方法:采用免疫组化法两步检测法检测正常乳腺组织、导管上皮不典型增生组织(ADH)以及浸润性导管癌(IDC)组织中的Skp2与p27Kip1的表达,并分析两者表达与乳腺癌淋巴结转移及组织学分级的关系。结果:Skp2阳性表达率在正常乳腺组织、ADH组织、IDC组织依次升高,伴腋窝淋巴结转移的IDC组织中Skp2阳性表达率明显高于无淋巴结转移的IDC组织,而p27Kip1的表达则与Skp2的表达呈反向变化(均P<0.05);不同组织学分级IDC组织间Skp2阳性率、p27Kip1阳性率差异均有统计学意义(均P<0.05),且Skp2阳性表达与IDC组织学分级呈正相关(r=0.492,P<0.05),而p27Kip1的阳性表达与IDC组织学分级呈负相关(r=-0.327,P<0.05)。结论:乳腺癌组织中Skp2表达上调而p27Kip1表达下调,两者的变化程度与乳腺癌的恶性生物学特征密切相关。
Objective: To investigate the expression of Skp2 and p27Kip1 in breast cancer and its significance. Methods: The expressions of Skp2 and p27Kip1 in normal breast tissue, ductal atypical hyperplasia (ADH) and invasive ductal carcinoma (IDC) tissues were detected by immunohistochemical method with two-step method. The expression of Skp2 and p27Kip1 in breast cancer Lymph node metastasis and histological grade relationship. Results: The positive expression of Skp2 in normal breast tissues, ADH tissues and IDC tissues increased in turn. The positive expression rate of Skp2 in IDC tissues with axillary lymph node metastasis was significantly higher than that in IDC tissues without lymph node metastasis, while the expression of p27Kip1 was correlated with the expression of Skp2 (P <0.05). The positive rates of Skp2 and p27Kip1 in different histological grade IDC tissues were significantly different (all P <0.05), and the positive expression of Skp2 was positively correlated with the histological grade of IDC (r = 0.492, P <0.05). However, the positive expression of p27Kip1 was negatively correlated with the histological grade of IDC (r = -0.327, P <0.05). Conclusion: Up-regulated Skp2 expression and down-regulated expression of p27Kip1 in breast cancer tissues are closely related to the malignant biological characteristics of breast cancer.
