目的探讨通用引物单链构象多态性技术用于呼吸道常见病原菌检测的可行性。方法选取7种常见呼吸道感染细菌,采用通用引物对细菌的16SrRNA基因进行扩增,对扩增产物进行单链构象多态性分析。结果经过扩增,7种常见病原菌均在300bp处有清晰条带,扩增产物的单链构象多态性图谱条带数为2~4条,相对迁移率及条带间距也存在明显不同,各细菌间可相互区分。结论通用引物SSCP技术能快速简便地鉴定呼吸道常见病原菌。
Objective To investigate the feasibility of universal primer single-strand conformation polymorphism (PCR) for the detection of common respiratory pathogens. Methods Seven common bacterial strains of respiratory tract infection were selected. The 16SrRNA gene of bacteria was amplified by universal primers, and single-strand conformation polymorphism analysis of the amplified products was carried out. Results After amplification, the seven common pathogenic bacteria showed a clear band at 300bp. The number of single strand conformation polymorphic bands of the amplified products was 2 to 4, and the relative mobility and band spacing were also significantly different. The bacteria can be distinguished from each other. Conclusion The common primer SSCP technique can identify common respiratory pathogens quickly and easily.
