目的:观察益气化浊胶囊对KKAy小鼠胰岛素抵抗及脂肪组织抵抗素(resistin)、视黄醇结合蛋白-4(RBP4)表达的影响。方法:将KKAy小鼠随机分为模型对照组(20 mL·kg-1·d-1)、益气化浊胶囊高、低剂量组(2,1 g·kg-1·d-1)、阳性对照(吡格列酮)组(2.5 mg·kg-1·d-1),另选C57BL/6J小鼠为空白对照组(20 mL·kg-1·d-1)。连续灌胃给药10周,每天1次,其中空白对照组、模型对照组,灌服无菌水。10周后测空腹血糖(FBG)、空腹血清胰岛素(FINS)并计算胰岛素敏感指数(ISI)和胰岛素抵抗指数(HOMA-IR)、脂肪组织抵抗素(resistin)、视黄醇结合蛋白(RBP)4含量。结果:模型对照组FBG,FINS,HOMA-IR,ISI,resistin,RBP4显著高于正常对照组(P<0.01);益气化浊高剂量组FBG低于模型对照组(P<0.05);益气化浊低、高剂量组FINS,HOMA-IR,ISI均低于模型组(P<0.01);益气化浊高剂量组resistin,RBP4低于模型组(P<0.05)。结论:模型组KKAy小鼠产生明显的胰岛素抵抗,表现为FBG,FINS,HOMA-IR,resistin,RBP4明显增高,ISI降低;益气化浊胶囊明显改善KKAy小鼠胰岛素抵抗状态,其结果为FBG,FINS,HOMA-IR,Resistin,RBP4降低,ISI升高,总体趋势为高剂量组优于低剂量组。
Objective: To observe the effect of Yiqi Huazhuo capsule on insulin resistance and expression of resistin and retinol binding protein-4 (RBP4) in KKAy mice. Methods: KKAy mice were randomly divided into model control group (20 mL · kg-1 · d-1), Yiqi Huazhuo Capsule high and low dose groups (2, 1 g · kg-1 · d-1) The positive control (pioglitazone) group (2.5 mg · kg-1 · d-1), and the other C57BL / 6J mice as the blank control group (20 mL · kg-1 · d-1). Administration of continuous gavage for 10 weeks, 1 day, including blank control group, model control group, sterile oral water. Fasting serum insulin (FINS) and fasting blood glucose (FBG) and insulin resistance index (HOMA-IR), resistin, retinol binding protein (RBP) 4 content. Results: FBG, FINS, HOMA-IR, ISI, resistin and RBP4 in the model control group were significantly higher than those in the normal control group (P <0.01); FBG in the high-dose Yiqi Huazhuo treatment group was lower than that in the model control group The levels of FINS, HOMA-IR and ISI in the low-dose and high-dose groups were lower than those in the untreated group (P <0.01). The resistin and RBP4 in the Yiqi Huazhuo high-dose group were lower than those in the model group (P <0.05). CONCLUSION: KKAy mice showed obvious insulin resistance in model group, showing FBG, FINS, HOMA-IR, resistin, RBP4 increased significantly, ISI decreased; Yiqikuaizhu capsule significantly improved insulin resistance in KKAy mice, the results for the FBG , FINS, HOMA-IR, Resistin, RBP4 decreased, ISI increased, the overall trend for the high-dose group was better than the low-dose group.
