目的分析浙江省肾综合征出血热(HFRS)疫区鼠形动物携带汉坦病毒状况及其型别。方法应用直接免疫荧光试验和RTPCR方法检测汉坦病毒抗原和核酸,阳性者用长爪沙鼠和VeroE6细胞进行病毒分离,对分离株的M片段基因进行序列测定和分析。结果 2008-2011年在浙江省HFRS疫区捕获的鼠形动物中以黑线姬鼠为优势鼠种;从该鼠鼠肺中共分离到6株汉坦病毒,序列分析表明分离株均为汉滩型汉坦病毒,其中Z521、524、534为H7亚型,TT-27、T-43、R88可能为新亚型;6株分离株之间的基因同源性较高,并与以往浙江省分离株具有较高同源性。结论 2008-2011年浙江省HFRS疫区鼠间存在汉滩型汉坦病毒流行。
Objective To analyze the status and type of hantavirus carried by mice in epidemic area of hemorrhagic fever with renal syndrome (HFRS) in Zhejiang province. Methods Direct immunofluorescence assay and RTPCR method were used to detect Hantavirus antigen and nucleic acid. The positive strains were isolated from gerbils with gerbils and Vero E6 cells. The M fragment gene of the isolate was sequenced and analyzed. RESULTS: From 2008 to 2011, Apodemus agrarius was the dominant species in the rodents captured in HFRS epidemic areas in Zhejiang Province. Six Hantaan viruses were isolated from the lungs of mice and the results showed that all the isolates were Hantan Hantaviruses, of which Z521, 524 and 534 were H7 subtypes, and TT-27, T-43 and R88 were probably new subtypes. The homologies among the 6 isolates were high, Isolates have high homology. Conclusions There is a Hantaan hantavirus epidemic in the HFRS-infected area of Zhejiang Province from 2008 to 2011.
