目的探讨姜黄素对体内人脑胶质瘤细胞U251生长的抑制作用。方法建立人脑胶质瘤细胞U251裸鼠皮下移植瘤模型后随机分为对照(C)组、姜黄素100mg/kg治疗(A)组和姜黄素50mg/kg治疗(B)组。腹腔注射给药30d,观察姜黄素治疗对移植瘤的体积抑制率和重量抑制率。治疗结束后收集各组皮下移植瘤组织,Westernblot法检测c-fos蛋白、细胞外调节蛋白激酶(ERK)、磷酸化细胞外调节蛋白激酶(p-ERK)表达。结果A、B两组移植瘤体积抑制率分别为62.4%、38.9%,重量抑制率分别为60.6%、36.4%。A、B两组肿瘤c-fos蛋白表达水平均明显低于C组,A组p-ERK蛋白水平明显低于C组。结论姜黄素对体内人脑胶质瘤细胞U251的生长有显著抑制作用。这可能与阻滞ERK信号通路有关。
Objective To investigate the inhibitory effect of curcumin on the growth of human glioma cells U251 in vivo. METHODS: Human glioma cells U251 were implanted into nude mice and subcutaneously implanted into nude mice and randomly divided into control (C), curcumin 100 mg/kg (A) and curcumin 50 mg/kg (B) groups. 30 days after intraperitoneal injection, curcumin therapy was used to observe the volume inhibition and weight inhibition of transplanted tumors. At the end of the treatment, all groups of subcutaneously transplanted tumor tissues were collected. The expression of c-fos protein, extracellular regulatory protein kinase (ERK) and phosphorylated extracellular regulatory protein kinase (p-ERK) was detected by Western blot. Results The volume inhibition rates of transplanted tumors in group A and B were 62.4% and 38.9%, and the weight inhibition rates were 60.6% and 36.4%, respectively. The expression levels of c-fos protein in both groups were significantly lower than those in group C. The levels of p-ERK protein in group A were significantly lower than those in group C. Conclusion Curcumin can significantly inhibit the growth of human glioma cells U251 in vivo. This may be related to blocking the ERK signaling pathway.
