试验以百日草雄性不育两用系AB201中的不育株和可育株为供试材料,采用BSA法和RAPD技术,在64个随机引物中筛选与育性基因紧密连锁的分子标记,结果发现引物BE-05在DNA纯合可育池和杂合可育池中扩增出一条长约1 500 bp的特异谱带,在DNA不育基因池中未发现特异谱带,该标记在3个育性不同的DNA池之间表现稳定的差异。用建池的单株DNA进一步对这个引物进行RAPD分析,发现仍表现稳定的多态性,该标记与育性基因MS紧密连锁,命名为BE-051500。
In this study, sterile and fertile plants of AB201, a male sterile line of Z. japonica, were used as experimental materials. Sixty-four random primers were screened for molecular markers closely linked to fertile genes using BSA and RAPD techniques. The results showed that the primer BE-05 amplified a specific band about 1 500 bp in DNA homozygous fertile and hybrid fertile cells and no specific band was found in DNA sterile gene pool, Stable differences were observed between the three fertile DNA pools. The RAPD analysis of this primer was further used to establish a pool of plant DNA and it was found that it still showed stable polymorphism. The marker was closely linked with the fertility gene MS and named as BE-051500.
