目的观察AGE对人脐静脉内皮细胞(HUVEC)标志物表达的影响。方法在体外培养的HUVEC细胞株中分别加入AGE、牛血清白蛋白(BSA),采用免疫组织化学技术和Western blot检测HUVEC中α-平滑肌肌动蛋白(α-SMA)和血管性血友病因子(vWF)的表达情况。结果 HUVEC经AGE处理7d后,AGE组α-SMA表达较BSA组增强[(27.04±0.05)vs(20.12±0.02),P<0.01],vWF表达较BSA组降低[(29.66±1.44)vs(55.28±1.77),P<0.01]。结论 AGE可体外诱导HUVEC表面标志物发生改变,该转分化机制可能参与糖尿病血管病变的发生发展。
Objective To observe the effect of AGE on the expression of human umbilical vein endothelial cells (HUVEC) markers. Methods AGE and bovine serum albumin (BSA) were respectively added into HUVEC cell lines cultured in vitro. The expression of α-smooth muscle actin (α-SMA) and vWF in HUVECs were detected by immunohistochemistry and Western blot (vWF) expression. Results Compared with BSA group, the expression of α-SMA in AGE group was significantly increased after AGE treatment for 7 days [(27.04 ± 0.05) vs (20.12 ± 0.02), P <0.01], and the expression of vWF in AGE group was lower than that in BSA group [(29.66 ± 1.44 vs 55.28 ± 1.77), P <0.01]. Conclusion AGE can induce the change of HUVEC surface markers in vitro, which may be involved in the development of diabetic vascular disease.
