目的探讨不同分期结肠癌患者血清及肿瘤组织中肿瘤坏死因子-α(tumor necrosis factor-α,TNF-α)和白细胞介素-6(interleukin-6,IL-6)的表达水平及其在结肠癌发病中的意义。方法 30例无肠道疾病患者为对照组,60例结肠癌患者为结肠癌组,其中Dukes分期A/B为早期,C/D为晚期。2组采用ELISA法检测血清TNF-α和IL-6水平,采用免疫组织化学法检测肿瘤患者癌旁组织、肿瘤组织中TNF-α和IL-6表达水平。结果结肠癌晚期患者血清TNF-α[(120.78±80.36)ng/L]、IL-6[(12.78±8.36)ng/L]高于结肠癌早期患者[(70.31±30.03)ng/L、(9.04±5.03)ng/L]和对照组[(10.92±10.01)ng/L、(2.94±1.39)ng/L](P<0.01),结肠癌早期患者高于对照组(P<0.01);早期和晚期结肠癌肿瘤组织TNF-α阳性表达率(43.3%、66.7%)、IL-6阳性表达率(76.7%、80.0%)均明显高于癌旁组织(TNF-α为3.3%、16.7%,IL-6为6.7%、10.0%)(P<0.05),晚期结肠癌肿瘤组织TNF-α、IL-6阳性表达率均高于早期(P<0.05);晚期结肠癌患者血清TNF-α和IL-6表达水平与肿瘤组织中表达水平呈正相关(r=0.65,P=0.00;r=0.52,P=0.00)。结论 TNF-α和IL-6在结肠癌发生、发展中起免疫调节作用,其血清水平检测有一定临床应用价值。
Objective To investigate the expression of tumor necrosis factor-α (TNF-α) and interleukin-6 (IL-6) in sera and tumor tissues of patients with different stages of colon cancer, The significance of cancer. Methods Thirty patients with no bowel disease were selected as the control group. Sixty colon cancer patients were selected as the colon cancer group. Dukes stage A / B was early stage and C / D stage was late stage. The serum levels of TNF-α and IL-6 were detected by ELISA in 2 and 2 groups. The expression of TNF-α and IL-6 in cancer tissues and adjacent tissues were detected by immunohistochemistry. The early stage of colon cancer was higher than that in the control group (P <0.01) The positive rates of TNF-α in early and advanced colon cancer tissues (43.3%, 66.7%) and IL-6 positive rates (76.7%, 80.0%) were significantly higher than those in paracancerous tissues (TNF- (P <0.05). The positive rates of TNF-α and IL-6 in advanced colon cancer tissues were higher than those in early stage (P <0.05) The expression of α and IL-6 was positively correlated with the expression of tumor tissue (r = 0.65, P = 0.00; r = 0.52, P = 0.00). Conclusion TNF-α and IL-6 play an immunomodulatory role in the development and progression of colon cancer. The detection of serum TNF-α and IL-6 has some clinical value.
