目的探讨邻苯二甲酸二(2-乙基己基)酯(diethylhexyl phthalate,DEHP)对小鼠血清及肺组织中谷胱甘肽过氧化物酶(GSH-Px)活力及谷胱甘肽(GSH)和过氧化氢含量的影响。方法将40只健康4周龄清洁级昆明小鼠按体重随机分为4组,分别为对照组和低(1/40 LD50,750 mg/kg)、中(1/20 LD50,1 500 mg/kg)、高(1/10 LD50,3 000 mg/kg)剂量DEHP染毒组,每组10只,雌雄各半。采用自由摄食方式进行染毒,连续染毒4周。测定小鼠血清和肺组织匀浆中GSH-Px活力和GSH、过氧化氢含量。结果 DEHP染毒可使小鼠血清和肺组织匀浆中GSH-Px活力和GSH含量显著下降,过氧化氢含量显著升高。结论 DEHP可致小鼠肺组织发生氧化性损伤,使机体的抗氧化能力降低。
Objective To investigate the effects of diethylhexyl phthalate (DEHP) on the activity of glutathione peroxidase (GSH-Px) and glutathione (GSH) in mice serum and lung tissue, And hydrogen peroxide content. Methods Forty healthy Kunming mice, aged 4 weeks, were randomly divided into 4 groups according to body weight: control group and low (1/40 LD50, 750 mg / kg), middle (1/20 LD50, 1500 mg / kg), high (1/10 LD50, 3 000 mg / kg) dose DEHP exposure group, each group 10, male and female. Adopt the free feeding way to carry on the poisoning, continuous poisoning 4 weeks. Determination of GSH-Px activity and GSH, hydrogen peroxide content in mouse serum and lung homogenate. Results DEHP exposure could significantly decrease GSH-Px activity and GSH content in serum and lung homogenate, and hydrogen peroxide content increased significantly. Conclusion DEHP can cause oxidative damage in the lungs of mice and reduce the body’s antioxidant capacity.
