目的:观察受体酪氨酸激酶TIE_2基因在脑血管畸形中的表达及突变。方法:57例脑血管病(46例脑AVMs)、36例正常人抽取外用静脉血6mL提取基因组 DNA;其中14例 AVMs显微外科手术切除AVM及AVM周围脑组织和8例正常脑血管标本,分别提取总RNA和4%PFA-PBS固定石蜡包埋。采用原位杂交、PCR-SSCP分析TIE_2受体在脑血管畸形中的变化。结果:部分AVMs和AVMs周围脑组织有受体酪氨酸激酶TIE_2 mRNA表达,在“动脉”和“静脉”之间没有明显差别;SSCP电泳结果显示2例 AVMs电泳迁移率与正常对照有差别,与受体酪氨酸激酶TIE_2阳性质粒DNA一致。结论:脑AVMs中有受体酪氨酸激酶TIE_2 mRNA表达,受体酪氨酸激酶TIE_2可能参与脑AVMs的发生。
Objective: To observe the expression and mutation of receptor tyrosine kinase TIE_2 gene in cerebrovascular malformations. Methods: Genomic DNA was extracted from 57 cases of cerebrovascular disease (46 cases of brain AVMs) and 36 cases of healthy volunteers. Sixteen AVM and AVM peripheral cerebral tissues and eight normal cerebrovascular samples were obtained by microsurgical resection of AVMs. Total RNA was extracted and embedded in 4% PFA-PBS fixed paraffin. In situ hybridization, PCR-SSCP analysis of TIE_2 receptor in cerebrovascular malformations. Results: The expression of TIE_2 mRNA in brain tissue around AVMs and AVMs did not show significant difference between “arteries” and “veins”. SSCP electrophoresis showed that the electrophoretic mobility shift of 2 AVMs was different from that of normal controls, Consistent with receptor tyrosine kinase TIE 2 positive plasmid DNA. CONCLUSION: The receptor tyrosine kinase TIE_2 mRNA is expressed in brain AVMs. The receptor tyrosine kinase TIE_2 may be involved in the pathogenesis of brain AVMs.
