目的建立高效液相色谱法同时测定豆豉片中栀子苷、大豆苷、染料木苷、大豆苷元、染料木素含量的方法。方法采用Hypersil ODS 2色谱柱(200 mm×4.6 mm,5-m);乙腈为流动相A,甲醇为流动相B,1.8%冰醋酸溶液为流动相C,洗脱程序为0 min(A:8%,B:10%,C:82%)→28 min(A:8%,B:10%,C:82%)→28.1 min(A:0%,B:38%,C:62%)→55 min(A:0%,B:38%,C:62%),流速为1 m L·min-1,检测波长为254 nm,柱温:30℃。结果栀子苷、大豆苷、染料木苷、大豆苷元、染料木素分别在12.36~396.00,3.50~112.00,3.36~116.00,3.00~96.00和1.63~52.00μg·m L-1内线性关系良好,平均回收率分别为100.0%,100.3%,99.8%,99.4%,99.9%,RSD分别为0.14%,0.97%,0.83%,0.98%,1.23%。结论本方法灵敏度高、专属性强、重复性好,可用于本制剂的质量控制。
Objective To establish a method for the simultaneous determination of geniposide, daidzin, genistin, daidzein and genistein by HPLC with high performance liquid chromatography (HPLC). Methods The chromatographic conditions were as follows: Hypersil ODS 2 (200 mm × 4.6 mm, 5-m); acetonitrile as mobile phase A, methanol as mobile phase B, 1.8% glacial acetic acid as mobile phase C, B: 10%, C: 82%) → 28 min (A: 8%, B: 10%, C: 82%) → 28.1 min (A: 0%, B: 38%, C: 62% ) → 55 min (A: 0%, B: 38%, C: 62%) at a flow rate of 1 m L · min-1 at a detection wavelength of 254 nm. Results Geniposide, daidzin, genistin, daidzein and genistein had good linearity in 12.36 ~ 396.00, 3.50 ~ 112.00, 3.36 ~ 116.00, 3.96 ~ 96.00 and 1.63 ~ 52.00 μg · m L-1, respectively The average recoveries were 100.0%, 100.3%, 99.8%, 99.4% and 99.9%, respectively. The RSDs were 0.14%, 0.97%, 0.83%, 0.98% and 1.23%, respectively. Conclusion The method has high sensitivity, specificity and repeatability, and can be used for the quality control of this preparation.
