目的 :检测miR-497/IGF-1R在肝癌中的表达并探讨其在肝癌侵袭与转移中的作用。方法 :定量PCR(real-time PCR)检测肝癌及癌旁组织中miR-497的表达,同时检测肝癌细胞系及正常人肝细胞中miR-497/IGF-1RmRNA的表达水平,免疫组织化学及Western blot检测肝癌与肝癌细胞系中IGF-1R蛋白表达,通过过表达或干扰肝癌细胞系中miR-497的表达,分析其对肝癌细胞侵袭与转移的作用。结果:肝癌组织中miR-497表达水平低于癌旁组织。与无脉管转移组相比,有脉管转移组降低更为明显。同时在肝癌细胞系也有相似的结果,高侵袭性肝癌细胞系MHCC-97H中降低最为明显。IGF-1R在肝癌组织及肝癌细胞系中表达增加。过表达miR-497能降低MHCC-97H中IGF-1R的表达,抑制其侵袭和转移;干扰miR-497表达能增加IGF-1R表达,促进SMMC-7721细胞的侵袭和转移。结论 :miR-497在肝癌组织中表达降低,过表达miR-497能下调IGF-1R,抑制肝癌的侵袭与转移,miR-497可能为治疗肝细胞肝癌提供新的靶点。
Objective: To detect the expression of miR-497 / IGF-1R in hepatocellular carcinoma (HCC) and to explore the role of miR-497 / IGF-1R in HCC invasion and metastasis. Methods: The expression of miR-497 in hepatocellular carcinoma and its adjacent tissues was detected by real-time PCR. The expression of miR-497 / IGF-1R mRNA in hepatocellular carcinoma cell lines and normal human hepatocytes was also detected. Immunohistochemistry and Western blot was used to detect the expression of IGF-1R protein in hepatocellular carcinoma and hepatocellular carcinoma cell lines. The expression of miR-497 in hepatocellular carcinoma cell lines was analyzed by Overexpression or Interference. Results: The expression of miR-497 in HCC tissues was lower than that in paracancerous tissues. Compared with no vascular transfer group, there is more obvious vascular transfer group. Similar results have also been observed in hepatocellular carcinoma cell lines, most significantly in the highly aggressive hepatocellular carcinoma cell line MHCC-97H. IGF-1R is expressed in hepatocellular carcinoma and hepatocellular carcinoma cell lines. Overexpression of miR-497 can reduce the expression of IGF-1R in MHCC-97H and inhibit its invasion and metastasis. Interfering with miR-497 can increase the expression of IGF-1R and promote the invasion and metastasis of SMMC-7721 cells. Conclusion: The expression of miR-497 is down-regulated in HCC. Overexpression of miR-497 downregulates IGF-1R and inhibits the invasion and metastasis of HCC. MiR-497 may provide a new target for the treatment of hepatocellular carcinoma.
