为研究ApoE基因对中国人非胰岛素依赖型糖尿病(NIDDM)患者血脂水平的影响,应用聚合酶链反应(PCR)扩增出ApoE基因第4外显子内的292bp片段,继以限制性内切酶HhaⅠ酶解消化,最后行8%聚丙烯酰胺凝胶电泳,根据电泳带型判断ApoE基因型。共检测了112例非胰岛素依赖型糖尿病患者的ApoE基因型。通过比较不同ApoE基因型与血脂水平的关系发现:ApoE基因多态性与TG,HDL-C,ApoAⅠ,ApoB及Lp(a)水平无相关性,而与TC(P=0.0029)及LDL-C(P=0.0021)水平相关,携带ε4等位基因者具有较高的TC及LDL-C水平,而携带ε2等位基因者具有较低的TC及LDL-C水平。
In order to study the effect of ApoE gene on blood lipid levels in Chinese patients with non-insulin dependent diabetes mellitus (NIDDM), a 292 bp fragment of ApoE gene exon 4 was amplified by polymerase chain reaction (PCR), followed by restriction endonuclease Enzyme Hha Ⅰ digestion, the final line of 8% polyacrylamide gel electrophoresis, ApoE genotypes according to the electrophoresis band. ApoE genotypes were detected in 112 non-insulin-dependent diabetic patients. By comparing the relationship between different ApoE genotypes and blood lipid levels, it was found that ApoE gene polymorphisms were not associated with TG, HDL-C, ApoAⅠ, ApoB and Lp (a) levels, but not with TC (P = 0.0029) C (P = 0.0021). The level of TC and LDL-C were higher in those with ε4 allele, while those with ε2 allele had lower TC and LDL-C levels.
