目的 :探讨运动对去势雌性大鼠骨组织BMP - 2 ,TNF -α表达的影响并讨论其在骨代谢变化中的意义。方法 :2 4只 4月龄雌性大鼠随机分为 3组 :对照组、单纯去势组、去势后运动组。单纯去势组和去势后运动组大鼠均行双侧卵巢切除。去势后运动组大鼠卵巢切除后在跑台上进行中等强度运动 (1 8m/min ,1h/day,5d/week) 1 0周。 1 0周后处死动物对血钙、血碱性磷酸酶、血骨钙素、尿钙、尿肌酐、尿羟脯氨酸进行检测以评定骨转换状态。同时对股骨近端松质骨中BMP - 2 ,TNF -α进行免疫组织化学染色 ,并结合计算机图像分析系统对表达强度进行评定。结果 :与对照组比较 ,单纯去势组大鼠骨吸收增强 ,骨转换加快 ,去势后运动组骨吸收和骨转换则无明显变化 ;与单纯去势组相比 ,去势后运动组大鼠骨转换和骨吸收均显著下调。单纯去势组大鼠TNF -α、BMP - 2阳性表达强度显著高于对照组 ,去势后运动组TNF -α、BMP - 2阳性表达与对照组相比无显著差异 ;去势后运动组大鼠骨组织中TNF-α、BMP - 2表达显著低于单纯去势组。结论 :运动对去势后大鼠的骨吸收增强和骨转换加快有抑制作用。卵巢切除后骨组织中TNF-α和BMP- 2高表达 ,运动对去势后骨组织中TNF -α和BMP - 2表达增加起下调作用
Objective: To investigate the effect of exercise on the expression of BMP - 2 and TNF - α in bone of ovariectomized female rats and its significance in the changes of bone metabolism. Methods: Twenty four 4-month-old female rats were randomly divided into three groups: control group, castrated group and castrated group. Bilateral ovariectomy was performed in both pure castration group and post-castration exercise group. After ovariectomized rats were ovariectomized, they were subjected to moderate intensity exercise (18 m / min, 1 h / day, 5 d / week) for 10 weeks on the treadmill. Animals were sacrificed at 10 weeks to detect serum calcium, blood alkaline phosphatase, serum osteocalcin, urinary calcium, urinary creatinine, and urinary hydroxyproline to assess bone turnover status. At the same time, immunohistochemical staining of BMP - 2 and TNF - α in the cancellous bone proximal to the femur was performed, and the expression intensity was evaluated by computer image analysis system. Results: Compared with the control group, the bone resorption and bone turnover were increased in the castration group and no significant changes in the bone resorption and bone turnover in the castrated group. Compared with the castrated group, the castrated group Rat bone turnover and bone resorption were significantly down-regulated. The positive expression of TNF - α and BMP - 2 in the castration group was significantly higher than that in the control group. There was no significant difference in the expression of TNF - α and BMP - 2 in the exercise group after castration compared with the control group. The expression of TNF - α and BMP - 2 in rat bone was significantly lower than that in simple castrate group. Conclusion: Exercise can inhibit the increase of bone resorption and bone turnover in castrated rats. After ovariectomy, the expression of TNF-α and BMP-2 in bone tissues were high, and exercise decreased the expression of TNF-α and BMP-2 in castrated bone
