目的 :探讨通脑精胶囊对胰岛素抵抗大鼠急性局灶性脑缺血的保护作用及其机制。方法 :高糖饲料喂养复制胰岛素抵抗 (IR)大鼠模型 ,再采用电凝大脑中动脉复制急性局灶性脑缺血动物模型 ,观察急性局灶性脑缺血IR大鼠一氧化氮 (NO)、一氧化氮合酶 (NOS)、超氧化物歧化酶(SOD)和肿瘤坏死因子α(TNFα)含量和活性的变化。结果 :通脑精胶囊显著降低缺血大鼠脑组织中NO含量和NOS活性及血浆TNFα的含量 (P <0 .0 1) ,增加脑组织中SOD活性 (P <0 .0 1)。结论 :通脑精胶囊对IR大鼠急性局灶性缺血有保护作用 ,其机制与降低NO、TNFα含量和NOS活性及增加SOD活性有关
Objective : To investigate the protective effect of Tongnaojing Capsule on acute focal cerebral ischemia in rats with insulin resistance and its mechanism. METHODS: Rats with insulin resistance (IR) were fed with high-sugar diet, and then the middle cerebral artery was used to replicate the animal model of acute focal cerebral ischemia. Nitric oxide (NO) was observed in IR rats induced by acute focal cerebral ischemia. ) Changes in nitric oxide synthase (NOS), superoxide dismutase (SOD), and tumor necrosis factor alpha (TNFα) content and activity. RESULTS: Tongnaojing Capsule significantly reduced NO content and NOS activity and plasma TNFα content (P <0.01), and increased SOD activity in brain tissue (P <0.01). Conclusion: Tongnaojing Capsule has protective effect on IR in rats with acute focal ischemia. The mechanism is related to the decrease of NO, TNFα content, NOS activity and increase of SOD activity.
