用甘油皮下注射复制兔急性肾小管坏死(ATN)模型,观察当归(RAS)对初发期ATN的防治效果,并初步探讨其作用机理。结果表明,注射甘油后24h,ATN组动物出现典型急性肾衰性肾功能改变和肾小管坏死.ATN+RAS组与ATN组比较,动物死亡率、滤过钠排泄分数和尿总氨基酸含量下降,血清肌酐、尿素氮和丙二醛的上升幅度减弱,肾组织钙和丙二醛含量降低,而超氧化物歧化酶、谷胱甘肽过氧化物酶、ATP酶活性升高,肾组织损伤程度也明显减轻。结果显示;当归对甘油所致ATN有明显防治作用,其机制可能与当归具有抑制脂质过氧化反应,保护肾脏抗氮化酶和ATP酶的活性,以及减轻肾组织钙超载有关
Acute renal tubular necrosis (ATN) was induced by subcutaneous injection of glycerol in rabbits. The preventive and therapeutic effects of Radix Angelicae Sinensis (RAS) on the onset of ATN were observed, and the mechanism of action was also discussed. The results showed that at 24 h after the injection of glycerol, ATN animals showed typical acute renal failure and renal tubular necrosis. ATN + RAS group compared with ATN group, animal mortality, sodium excretion fraction and total urinary amino acid content decreased, the serum creatinine, urea nitrogen and malondialdehyde increased less, the content of calcium and malondialdehyde decreased, while the superoxide Dismutase, glutathione peroxidase, ATPase activity increased, the extent of renal tissue damage also significantly reduced. The results showed that Angelica sinensis on ATN caused by glycerol has a significant preventive effect, and its mechanism may be related to Angelica inhibition of lipid peroxidation, protection of renal activity of nitrilase and ATPase, as well as to reduce renal calcium overload
