目的:探讨十全大补汤对Lewis肺癌组织中MMP-9基因表达的影响。方法:50只小鼠采用细胞株接种的方法复制Lewis肺癌转移模型,造模成功后,将小鼠分为模型组,CTX组,十全大补汤低、中、高剂量组。采用RT-PCR方法检测十全大补汤对Lewis肺癌组织中MMP-9基因表达的影响。结果:十全大补汤中、高剂量组的肿瘤转移抑制率为43.20%、29.59%,两组中MMP-9的表达下降,与模型组比较有统计学意义(P<0.01)。结论:十全大补汤下调MMP-9基因表达是其抑制肺癌转移的分子机制之一。
Objective: To investigate the effect of Shiquan Dabu Tang on the expression of MMP-9 in Lewis lung carcinoma. Methods: Fifty mice were used to inoculate Lewis lung carcinoma metastasis model by cell inoculation method. After successful modeling, the mice were divided into model group, CTX group and Shiquandabu Tang low, middle and high dose group. The effect of Shiquan Dabu Tang on MMP-9 gene expression in Lewis lung cancer tissues was detected by RT-PCR. Results: The inhibition rate of metastasis in Shiquandupu Decoction group was 43.20% and 29.59%, respectively. The expression of MMP-9 in both groups was significantly lower than that in model group (P <0.01). Conclusion: Shiquan Dabu Tang down-regulates the expression of MMP-9 gene is one of the molecular mechanisms of its inhibition of lung cancer metastasis.
