目的:观察葡萄糖转运体1(GLUT1)在妊娠期糖尿病(GDM)大鼠胎盘中的表达水平。方法:选取14例雌性SD大鼠,腹腔注射链脲菌素,65 mg/kg,3天后测定大鼠血糖水平,超过250 mg/dL即为成功的GDM大鼠模型。GDM大鼠与正常雌性大鼠合笼交配,观察阴道涂片,记录妊娠日期。提取胎盘组织RNA,应用RT-PCR和实时PCR观察妊娠第21天大鼠胎盘GLUT1的表达水平。结果:与正常妊娠大鼠比较,GDM大鼠胎盘GLUT1表达水平降低。结论:GDM大鼠胎盘GLUT1的表达水平降低。
Objective: To observe the expression of glucose transporter 1 (GLUT1) in the placenta of gestational diabetes mellitus (GDM) rats. Methods: Fourteen female Sprague-Dawley rats were injected intraperitoneally with streptozotocin at a dose of 65 mg / kg. The blood glucose level was measured after 3 days. A blood glucose level above 250 mg / dL was considered as a successful GDM rat model. GDM rats and normal female rats caged mating, vaginal smear observed, record the date of pregnancy. The placental tissue RNA was extracted, and the expression of GLUT1 in placenta of the 21st day of pregnancy was detected by RT-PCR and real-time PCR. Results: Compared with normal pregnant rats, the expression of GLUT1 in placenta of GDM rats decreased. Conclusion: The expression of GLUT1 in placenta of GDM rats decreased.
