本文采用固定细胞ELISN,间接免疫光和SPA花环三种方法同时检测抗白血病细胞表面抗原单克隆抗体.结果证明固定细胞ELISA法阳性率最高.SPA花环法较低,间接免疫荧光法是筛选细胞表面单抗的最好方法. 采用双抗体夹心法检测杂交瘤上悬液中免疫球蛋白分泌情况。结果提示双抗体夹心法阳性者用三种筛选方法证明可为阴性.提示这种抗体可能是非特异性抗体.不与细胞表面抗原反应。
In this paper, monoclonal antibody against leukemia cell surface antigen was simultaneously detected by three methods of fixed cell ELISN, indirect immunofluorescence and SPA wreath. The results showed that the positive rate of fixed cell ELISA was the highest, the SPA rosette method was low, indirect immunofluorescence method was used to screen the cell surface Monoclonal antibody best method.Using the double antibody sandwich method to detect hybridoma tumor suspension immunoglobulin secretion. The results suggest that double antibody sandwich positive method with three screening methods can be proved to be negative, suggesting that this antibody may be non-specific antibodies do not react with cell surface antigens.
