过去曾应用许多流感病毒株进行过RNA多聚酶活性的研究,但是都没有肯定所用的反应条件对不同的毒株是否最适。在本文中,作者比较了5个流感病毒株(其中包括4个A型流感亚型病毒):A/PRg/34(H_0N_1),A/WS/33 (H_0 N_1),A/FM_1/47 (H_1 N_1),A/Singa-pore/57(H_2 N_2),和Victoria/75(H_3 N_2)病毒的RNA多聚酶的活性。观察了培养温度以及对热的稳定性。按照Pons和Hirst的方法进行病毒的提纯浓缩。根据Bishop等所推荐的方法测定酶的活性,用闪烁分光光度计测定放射性。
In the past, many influenza virus strains have been used to study the activity of RNA polymerase. However, it is not certain whether the reaction conditions used are optimal for different strains. In this paper, we compared five influenza virus strains, including four influenza A subtypes: A / PRg / 34 (H_ON_1), A / WS / 33 (H_0N_1), A / FM_1 / 47 H_1 N_1), A / Singa-pore / 57 (H_2 N_2), and Victoria / 75 (H_3 N_2) viruses. The incubation temperature and the stability to heat were observed. The virus was purified and concentrated according to the method of Pons and Hirst. The activity of the enzyme was determined according to the method recommended by Bishop et al., And the radioactivity was measured with a scintillation spectrophotometer.
