目的 探讨脑细胞生长肽对缺氧缺血新生鼠 (HIBD)脑的保护作用。方法 7日龄Wistar大鼠 ,72只 ,随机分成 3组 ,(1)假手术组 (n =12 ) ;(2 )观察组 (n =30 ) :HIBD模型后即刻给脑细胞生长肽 10mg腹腔注射 ;(3)对照组 (n =30 )HIBD模型后即刻腹腔注射等体积的生理盐水。于处置后 0、2 4、4 8、72h检测脑组织中超氧化物歧化酶 (SOD)、丙二醛 (MDA)和一氧化氮 (NO)水平 ;并于处置后 2 4h作HE和Tunel染色光镜下检测脑细胞凋亡数。结果 观察组脑组织SOD、MDA、NO水平及脑细胞凋亡数与对照组比较均有显著性差异。结论 脑细胞生长肽可通过拮抗氧自由基、抑制NO合成和阻止神经细胞凋亡 ,发挥对HIBD时脑的保护作用
Objective To investigate the protective effect of brain growth peptide on brain of hypoxic-ischemic neonatal rats (HIBD). Methods Seven-day-old Wistar rats, 72, were randomly divided into 3 groups: (1) sham operation group (n = 12); (2) observation group (n = 30) Injection; (3) control group (n = 30) immediately after HIBD model intraperitoneal injection of equal volume of saline. The levels of superoxide dismutase (SOD), malondialdehyde (MDA) and nitric oxide (NO) were detected at 0,2,4,48,72h after treatment. HE and Tunel staining Light microscope to detect the number of apoptotic brain cells. Results The levels of SOD, MDA, NO in brain tissue and the number of apoptotic cells in observation group were significantly different from those in control group. Conclusion Brain cell growth peptide can protect the brain in HIBD by antagonizing oxygen free radicals, inhibiting NO synthesis and preventing neuronal apoptosis
