目的探讨SAHA(suberoylanilide hydroxamic acid)和肿瘤坏死因子相关凋亡诱导配体(TNF-related apoptosis inducing ligand,TRAIL)联合作用抑制雌激素受体阳性乳腺癌细胞MCF-7生长的分子机制。方法以乳腺癌细胞株MCF-7为研究对象,应用自动细胞分析仪测定SAHA和TRAIL联合作用对乳腺癌细胞活力和细胞凋亡的影响并对细胞周期进行分析;应用实时定量PCR及固相凋亡抗体芯片检测MCF-7细胞凋亡蛋白表达。结果 SAHA和TRAIL联合作用可显著降低MCF-7细胞活力,抑制细胞增殖,并诱导细胞凋亡的产生。SAHA和TRAIL对MCF-7细胞周期的影响主要集中在G_0/G_1期。结论 SAHA和TRAIL联合作用对乳腺癌细胞生长有抑制作用。
Objective To investigate the molecular mechanism of SAHA (suberoylanilide hydroxamic acid) and tumor necrosis factor-related apoptosis inducing ligand (TRAIL) in inhibiting the growth of estrogen receptor-positive breast cancer cell line MCF-7. Methods The breast cancer cell line MCF-7 was used as the research object. The effect of the combination of SAHA and TRAIL on the cell viability and apoptosis of breast cancer cells was assayed by automatic cell analyzer. The cell cycle was analyzed. Real- Detect the expression of apoptosis protein in MCF-7 cells by using antibody. Results The combination of SAHA and TRAIL could significantly decrease the viability of MCF-7 cells, inhibit cell proliferation and induce apoptosis. The effects of SAHA and TRAIL on the cell cycle of MCF-7 mainly concentrated on the G_0 / G_1 phase. Conclusion The combination of SAHA and TRAIL can inhibit the growth of breast cancer cells.
