为研究贮藏条件和品种因素对马铃薯块茎中α-茄碱含量的影响,本试验建立了利用超高效液相色谱-三重四级杆串联质谱(UPLC-QQQ)测定马铃薯块茎中α-茄碱含量的方法,通过正交试验研究了5个品种的马铃薯块茎在不同贮藏温度、光照强度、贮藏时间下α-茄碱含量的变化情况。结果表明,该检测方法检测时间短,灵敏度高,重复性好。5种马铃薯块茎中的α-茄碱含量均随3种贮藏因素水平的增大而增加;方差分析表明,3种因素影响作用的主次顺序为光照强度>贮藏时间>贮藏温度。因此,在低温避光的条件下能够较好地控制马铃薯块茎中α-茄碱的产生。冀张8号品种的α-茄碱增长速度较快,且其受贮藏时间因素的影响较大,在控制α-茄碱方面属于不宜长期贮藏品种。本研究结果为马铃薯块茎贮藏过程中α-茄碱的含量控制提供了理论参考。
In order to study the effect of storage conditions and variety factors on the content of α-solanine in potato tubers, we established the method of UPLC-QQQ The orthogonal test was used to study the changes of α-solanine content under different storage temperature, light intensity and storage time for five varieties of potato tubers. The results show that the detection method has short detection time, high sensitivity and good repeatability. The contents of α-solanine in five kinds of potato tubers increased with the increase of three storage factors. The analysis of variance showed that the main order of the three kinds of factors was light intensity> storage time> storage temperature. Therefore, under low temperature and dark conditions, the production of α-solanine in potato tubers can be well controlled. Ji Zhang 8 varieties of α-solanine growth rate faster, and its storage time factors, the larger, in the control of α-solanine are not suitable for long-term storage of varieties. The results of this study provide a theoretical reference for the control of α-solanine in potato tubers.
