肿瘤和艾滋病 (AIDS)的基因治疗或免疫治疗的研究需要大量的T细胞克隆。尽管T细胞来源于CD34+细胞 ,但由于需要胸腺组织的参与 ,使其在体外扩增大量细胞克隆非常困难。本研究用脐血单个核细胞作为辅助细胞 ,在SCF +IL 2等细胞因子的作用下 ,人脐血CD34+在此培养条件下 ,不断产生CD4 +或CD8+T细胞至少持续 3周。在培养扩增过程中 ,发现CD4 +和CD8+T细胞的比例有一个不断变化的动态过程 ,培养体系中可以检测到CD4和CD8双阳性细胞的存在 ;RT PCR可检测到负责TCR重排的RAG 2基因的表达 ,说明所获得的T细胞来源于CD34+细胞。本研究为从CD34+细胞获得大量的T细胞克隆提供了一个简便的体外培育体系。
Studies of gene therapy or immunotherapy of tumors and AIDS (AIDS) require large numbers of T cell clones. Although T cells are derived from CD34 + cells, it is very difficult to expand a large number of cell clones in vitro due to the need of thymus tissue. In this study, umbilical cord blood mononuclear cells as helper cells, in the SCF + IL 2 and other cytokines, human umbilical cord blood CD34 + in this culture conditions, continue to produce CD4 + or CD8 + T cells for at least 3 weeks. During the course of culture and expansion, we found that the proportion of CD4 + and CD8 + T cells has a dynamic process that can detect the presence of both CD4 and CD8 double positive cells in culture system. RT PCR can detect the TCR rearrangement RAG 2 gene expression, indicating that the T cells obtained from CD34 + cells. This study provides a convenient in vitro culture system for obtaining large numbers of T cell clones from CD34 + cells.
