目的依据生育三烯酚的抗肿瘤作用,研究其对结肠癌细胞SW620细胞增殖抑制作用与Wnt信号途径之间的关系。方法采用MTT方法,观察不同剂量δ-生育三烯酚对细胞增殖抑制作用,通过Real-time RT-PCR检测Wnt途径相关因子mRNA的表达量变化情况,观察δ-生育三烯酚对细胞中wnt-1、β-catenin、c-jun、c-myc、cyclin D1及MMP-7mRNA表达水平的影响。结果δ-生育三烯酚对结肠癌SW620细胞增殖有明显抑制作用,20μmol/Lδ-生育三烯酚作用24 h对SW620细胞抑制率为70.43%,IC50值为15.18μmol/L。20μmol/Lδ-生育三烯酚对Wnt信号途径中的wnt-1、β-catenin、c-jun及c-myc mRNA表达降低(P<0.05);对cyclin D1及MMP-7 mRNA的表达无影响(P>0.05)。结论δ-生育三烯酚对结肠癌SW620细胞增殖抑制作用可能与其致Wnt途径中wnt-1、β-catenin、c-jun及c-myc表达降低有关。
OBJECTIVE To study the antitumor effect of tocotrienol on the proliferation of human colon cancer cell line SW620 and its relationship with Wnt signaling pathway. Methods MTT assay was used to observe the effects of different doses of δ-tocotrienol on cell proliferation. Real-time RT-PCR was used to detect the mRNA expression of Wnt pathway-related factors. The effects of δ-tocotrienol on the expression of wnt 1, β-catenin, c-jun, c-myc, cyclin D1 and MMP-7mRNA expression levels. Results δ-Tocotrienol had a significant inhibitory effect on the proliferation of SW620 colon cancer cells. The inhibition rate of SW620 cells treated with 20μmol / L δ-Tocotrienol for 24 h was 70.43% and the IC50 was 15.18μmol / L. The expression of wnt-1, β-catenin, c-jun and c-myc mRNA in Wnt signaling pathway was decreased in 20μmol / L δ-tocotrienol treatment group (P <0.05), but had no effect on the expression of cyclin D1 and MMP- (P> 0.05). Conclusion The inhibitory effect of δ-tocotrienol on the proliferation of colon cancer SW620 cells may be related to the decrease of Wnt-1, β-catenin, c-jun and c-myc expression in Wnt pathway.
