目的: 建立一种高效液相色谱快速测定人血清中头孢曲松浓度的分析方法。方法: 使用Bio- Rad 高效液相色谱仪, LunaTM C18 (2) 柱(4-6 m m ×150 m m , 5 μm) , 流动相为甲醇- 0-1 % 三乙胺水溶液(15∶85 , 用磷酸调pH 至7-5) , 检测波长为240 nm , 内标为茶碱。结果: 头孢曲松人血清浓度在0-75 ~384 μg·m L- 1 范围内线性良好, 最低检出浓度为0-5 μg·m L- 1 , 日内及日间精密度均优于5 % 。结论: 本方法经济、快速、灵敏、准确、专属性高。
Objective: To establish a method for the rapid determination of ceftriaxone in human serum by high performance liquid chromatography. Methods: Bio-Rad high performance liquid chromatography (LunaTM C18 (2) column (4-6 m × 150 m m, 5 μm) was used with the mobile phase consisted of methanol and 0-1% , PH adjusted to 7-5 with phosphoric acid), detection wavelength was 240 nm, the internal standard for theophylline. Results: The linearity of ceftriaxone in the range of 0-75 ~ 384 μg · m L -1 was linear with the lowest detectable concentration was 0-5 μg · m L -1. The intra- and inter-day precision was better than 5 %. Conclusion: The method is economical, fast, sensitive, accurate and specific.
