目的 建立测定盐酸托烷司琼的人体血药浓度的方法 ,用于其血药浓度测定并进行临床药代动力学研究。方法 采用高效液相 -二极管阵列色谱法测定盐酸托烷司琼的血药浓度。结果 盐酸托烷司琼的校正标准曲线方程为 ^Y= - 0 0 35 7+0 0 1 7X(r=0 990 9) ,其 5 ,1 0 ,2 5 μg·L-1 3浓度的血样回收率分别为 1 0 3 2 4 % ,97 5 1 % ,1 0 4 95 % ;精密度分别 5 6 7% ,7 83% ,8 1 8% ;志愿受试者禁食口服 2 0mg盐酸托烷司琼胶囊后 ,盐酸托烷司琼的Cmax为 2 7 4 4 μg·L-1 。结论 此方法可满足测定要求 ,可用于盐酸托烷司琼的药代动力学研究
OBJECTIVE To establish a method for the determination of tropisetron hydrochloride in human plasma, and to study its pharmacokinetics in plasma. Methods The plasma concentration of tropisetron hydrochloride was determined by high performance liquid chromatography with diode array. Results The standard curve equation of tropisetron hydrochloride was as follows: Y = - 0 0 35 7 + 0 0 1 7X (r = 0 990 9), and the blood concentration of 5, 10, 25 μg · L -1 3 The recovery rates were 103.24%, 97.51% and 104.4%, respectively. The precision were 56.7%, 73.3% and 81.8% respectively. The volunteers were fasted and fed with 20 mg HCl After fentanyl capsule, the Cmax of tropisetron hydrochloride was 2 744 μg · L -1. Conclusion This method can meet the determination requirements, can be used for the pharmacokinetics of tropisetron hydrochloride
