本文建立了提取与测定大鼠腹腔巨噬细胞(PMφ)生物合成白三烯B_4(LTB_4)及5-羟基二十碳四烯酸(5-HETE)的高效液相色谱法,并研究了4个单味中药抗炎作用与对PMφ产生LTB_4及5-HETE的影响。结果表明大黄、厚朴、白头翁及败酱草提取物对大鼠PMφ合成以上二种介质均有明显抑制作用。对炎性介质LTB_4产生的抑制率(%)分别为:大黄素(终浓度取5.0×10~(-6)mol/L)为88.0%;厚朴为99.8%;白头翁为94.9%及败酱草为57.9%(以上终浓度均取5.9×10~(-2)g/L)。
In this paper, a high performance liquid chromatography method for the extraction and determination of rat peritoneal macrophage (PMφ) biosynthetic leukotriene B_4 (LTB_4) and 5-hydroxyeicosatetraenoic acid (5-HETE) was established and studied. The anti-inflammatory effects of single Chinese herbs and their effects on PMΦ production of LTB_4 and 5-HETE. The results showed that extracts of Rhubarb, Magnolia officinalis, Pulsatilla chinensis, and Patriniae extract had significant inhibitory effects on the two media of PMφ synthesis in rats. The inhibition rate (%) of inflammatory mediators LTB_4 was 88.0% for emodin (final concentration 5.0×10 −6 mol/L), and 99.8% for Magnolia; Pulsatilla It was 94.9% and 57.9% of P. edulis (above the final concentration was 5.9×10-2 g/L).
