目的研究竹节蓼体外抗肿瘤活性及确定其活性部位。方法采用MTT法和细胞形态观察法研究竹节蓼醇提物及醇提物的4个不同提取部位对4种肿瘤细胞株A549、PC-3、SGC-7901、Hep G2增殖的抑制作用及形态的影响,确定抗肿瘤活性部位;采用DAPI染色法研究乙酸乙酯部位对A549细胞凋亡形态学的影响。结果竹节蓼醇提物、竹节蓼乙酸乙酯和石油醚提取部位对上述4种肿瘤细胞的生长均有不同程度的抑制作用且具有剂量依赖关系,且均能显著改变细胞形态;DAPI染色结果显示乙酸乙酯部位促进了A549细胞凋亡。结论竹节蓼中具有抗肿瘤作用的活性物质,有效成分集中在乙酸乙酯和石油醚提取部位。
Objective To study the antitumor activity of Polygonum viviparum in vitro and its active site. Methods MTT assay and cell morphology observation were used to study the inhibitory effects and the morphological changes of four different tumor cell lines A549, PC-3, SGC-7901 and Hep G2 To determine the anti-tumor active site; DAPI staining was used to study the influence of ethyl acetate fraction on apoptosis morphology of A549 cells. Results Alcohol extract of Polygonum cuspidatum and ethyl acetate extract of Polygonum cuspidatum and petroleum ether had different degrees of inhibition on the growth of the above - mentioned 4 kinds of tumor cells in a dose - dependent manner, and all of them could significantly change the cell morphology. DAPI staining The results showed that the ethyl acetate fraction promoted the apoptosis of A549 cells. Conclusion Polygonum nobile has anti-tumor activity of active substances, the active ingredient concentrated in ethyl acetate and petroleum ether extraction site.
