目的探讨大肠癌组织中HSP70和p53蛋白的表达特点及其相互间关系。方法运用SP法结合图像分析,探讨HSP70和p53蛋白在大肠癌组织、癌旁组织及正常组织中的表达。结果在31例大肠癌、癌旁、正常组织标本中,HSP70表达的平均光密度值分别为:0.5917±0.0678、0.3363±0.0485、0.1742±0.0193;癌组织中HSP70的表达水平显著升高,与癌旁、正常组织中的表达比较差异有统计学意义(P<0.01)。p53蛋白表达的平均光密度值分别为:0.7547±0.2732、0.5388±0.1542、0.3770±0.0682;癌组织中p53蛋白的表达水平显著升高,与癌旁、正常组织中的表达比较差异有统计学意义(P<0.01)。HSP70和p53在大肠癌组织中的表达存在依从关系(P<0.01),二者呈直线相关,相关系数r为0.795。结论HSP70和p53蛋白在大肠癌组织中高表达,提示二者在大肠癌的发生发展中起一定的作用,是细胞恶变的指标之一;他们在大肠癌的发生发展中具有协同作用,或者在大肠癌组织中可能存在着HSP70-p53蛋白的复合物形式。
Objective To investigate the expression of HSP70 and p53 protein in colorectal carcinoma and its relationship with each other. Methods SP method combined with image analysis to explore the expression of HSP70 and p53 protein in colorectal cancer tissues, adjacent tissues and normal tissues. Results The average optical density of HSP70 expression in 31 cases of colorectal cancer, adjacent normal tissue and normal tissue were 0.5917 ± 0.0678,0.3363 ± 0.0485,0.1742 ± 0.0193 respectively. The expression of HSP70 in cancerous tissue was significantly increased, There were significant differences in the expression of P53 in normal tissues (P <0.01). The average optical density of p53 protein expression was 0.7547 ± 0.2732,0.5388 ± 0.1542,0.3770 ± 0.0682 respectively. The expression of p53 protein in cancer tissues was significantly higher than that in adjacent non-cancerous and normal tissues (P <0.01). The expression of HSP70 and p53 in colorectal cancer tissues was in a compliance relationship (P <0.01), and there was a linear correlation between the two. The correlation coefficient r was 0.795. Conclusions High expression of HSP70 and p53 protein in colorectal cancer suggests that both of them play a role in the carcinogenesis and progression of colorectal cancer and is one of the indicators of cell malignant transformation. They have synergistic effect in the development of colorectal cancer, or in the large intestine Cancer tissue may exist in the HSP70-p53 protein complex forms.
