目的:探讨健脑益智方对脑缺血再灌注损伤后炎性细胞因子肿瘤坏死因子-α(TNF-α)、白细胞介素-1β(IL-1β)含量的影响。方法:健康SD大鼠随机分为5组,每组6只,分别为假手术组、模型组、中药干预后造模组、阿司匹林干预后造模组、中药治疗组。制备大鼠全脑缺血再灌注模型,于全脑缺血20min,再灌注24h后,测定血清及脑组织中TNF-α和IL-1β的含量。结果:模型组大鼠血清及脑组织中TNF-α、IL-1β的含量高于假手术组(P<0.01),阿司匹林干预后造模组、中药干预后造模组及中药治疗组TNF-α和IL-1β的含量均低于模型组(P<0.01,P<0.05)。结论:健脑益智方可降低炎性细胞因子TNF-α、IL-1β的含量,并可能通过此途径拮抗脑缺血再灌注炎症损伤反应,从而起到脑保护作用。
Objective: To investigate the effects of Jiannao Yizhi Recipe on the levels of inflammatory cytokines TNF-α and IL-1β after cerebral ischemia-reperfusion injury. Methods: Healthy Sprague-Dawley rats were randomly divided into 5 groups (6 rats in each group), which were sham-operation group, model group, model group after TCM intervention, model group after aspirin intervention, and TCM treatment group. The model of global cerebral ischemia-reperfusion in rats was established. The contents of TNF-α and IL-1β in serum and brain tissues were determined after 20 min of global cerebral ischemia and 24 h of reperfusion. Results: The contents of TNF-α and IL-1β in serum and brain tissues of model group were higher than those of sham operation group (P <0.01). After intervention of aspirin, α and IL-1β were lower than the model group (P <0.01, P <0.05). Conclusion: Jiannaoyizhi can reduce the levels of inflammatory cytokines TNF-α, IL-1β, and may antagonize the inflammatory injury induced by cerebral ischemia-reperfusion through this pathway, which may play a role in cerebral protection.
