为了探讨西芹根物质中化感物质对黄瓜枯萎病菌生理生化的作用,本研究对西芹根物质丙酮浸提液进行4次柱层析,每次层析后均进行化感效果测定,以化感抑制效果筛选每次层析最佳流分;然后将最佳流分和黄瓜枯萎病菌在液体培养基条件下进行共培养,144 h后测定黄瓜枯萎病菌分泌的3种细胞壁降解酶活性以及镰刀菌酸的含量。结果表明:西芹鲜根及根际土丙酮浸提液4次层析物每次层析获得的最佳流分均能抑制黄瓜枯萎病菌的生长;随层析次数的增加细胞壁降解酶活性和镰刀菌酸含量呈下降趋势,且细胞壁降解酶活性和镰刀菌酸含量较对照差异显著;相关性分析结果显示,各层析物化感抑制效果与果胶酶和β-葡萄糖苷酶的活性呈显著负相关,与镰刀菌酸含量无显著相关关系。
