目的:建立反相高效液相色谱法测定黄芩漱口液中黄芩苷的含量。方法:采用反相高效液相色谱法,以依利特Krosmasil-C18柱(4.6mm×250mm,5μm)为色谱柱,甲醇–0.2%磷酸溶液(48∶52)为流动相,流速1.0mL·min-1,检测波长280nm,进样量10μL,柱温30℃。结果:在给定的色谱条件下,黄芩苷与相邻组分峰之间分离良好,阴性对照试验无干扰。黄芩苷浓度在20.0~200.0μg·mL-1之间与峰面积呈良好线性关系,回归方程Y=0.4937X+0.3041,r=1.000,平均加样回收率(n=6)为99.0%;RSD=1.73%。结论:本法操作简便,专属性好,结果准确,可作为黄芩漱口液的含量控制方法。
Objective: To establish a method for the determination of baicalin in baicalin gargle by reversed-phase high performance liquid chromatography. Methods: The elution Krosmasil-C18 column (4.6mm × 250mm, 5μm) was used as the mobile phase in the mobile phase of methanol-0.2% phosphoric acid solution (48:52) at a flow rate of 1.0mL · min -1, detection wavelength 280nm, injection volume 10μL, column temperature 30 ℃. Results: Under the given chromatographic conditions, the separation between baicalin and adjacent component peaks was good, and the negative control experiment had no interference. The concentration of baicalin showed a good linear relationship with the peak area in the range of 20.0 ~ 200.0μg · mL-1. The regression equation was Y = 0.4937X + 0.3041, r = 1.000, and the average recovery was 99.0% (n = 6) = 1.73%. Conclusion: This method is simple and convenient to operate, with good specificity and accurate results. It can be used as a method to control the content of Scutellariae gargle.
